Gene Autry hunts bank robbers Al Bartlett and Trot Lucas with his old friend Mike. Bartlett, to throw off his pursuers, kills Trot and his own brother. When Kitty Bartlett comes to town claiming to be the slain Bartlett's widow, Gene has to save her from the irate townspeople who are not aware that her name isn't Bartlett but she really is the daughter of a law officer slain by Al Bartlett. Ben Luder, a local hood, tricks Bartlett back into town by saying he has to fixed to have Doc Larry Taylor do plastic surgery on him. En route they meet Doc and his assistant Helen Ellis and Ben's ruse is exposed. Bartlett kills Ben and forces Doc to drive him to the railroad. Gene, in a fight atop a runaway train, captures Bartlett.
Three Union POWs fleeing across the desert to escape both their Confederate pursuers and rampaging Apaches come across a dying woman and her infant child. They promise the woman that they will take care of the child and get it to safety, even though none of them knows anything whatsoever about children or babies.

The film starts off with an old man in the desert and two signs are shown; one that says East, the other says west. Japan has sent a mission to San Francisco. In San Francisco, the Japanese are surprised by the American culture. One American, Gus Taylor, and his gang steal all the gold from the mission and make their way in to the desert. One of the samurai of the mission chases after the gang into the desert. He is joined by a young American boy, Sam, whose father was killed by the gang leader. The group picks up a variety of people along the way to New Mexico. The Japanese and the Americans on the trip share parts of their own cultures with each other. The group of vigilantes eventually makes it to New Mexico and finds the gang. They take back their stolen gold and return to San Francisco.
Keaton portrays Friendless, who travels west to try to make his fortune. Once there, he tries his hand at bronco-busting, cattle wrangling, and dairy farming, eventually forming a bond with a cow named "Brown Eyes." Eventually he finds himself leading a herd of cattle through Los Angeles.
